Abstract
|
L-Asparaginase (L-asparagine amidohydrolase EC 3. 5. 1. 1) catalyses the hydrolysis of L-asparagine to L-aspartic acid and ammonia. L-Asparaginase is considered for its therapeutic properties and industrial applications. The aim of this study was optimization of the L-Asparaginase production in the yeast Yarrowia lipolytica using statistical modeling method. Levels of four main factors glucose, glutamine, potassium hydroxide and pH were optimized by Taguchi experimental design method to improvement of L-Asparaginase production in Y. lipolytica DSM3286. After optimization, L-Asparaginase production was reached to 62 U/mL which is about 4 times more than primary medium. Therefore, the Taguchi experimental design method is useful method for statistical optimization and cost-effective production of L-Asparaginase in Y. lipolytica.
