چکیده Background: Laccases (benzenediol:oxygen oxidoreductases, EC 1.10.3.2) are copper-containing polyphenol oxidases that can oxidize a wide range of aromatic and phenolic compounds. They have good potential for industrial applications and for reducing environmental pollution. The aim of this study was to investigate the effect of different substrates on heterologous production of a Trametes versicolor laccase in a recombinant strain of the non-conventional yeast Yarrowia lipolytica. Materials and Methods: The recombinant Y. lipolytica YL4 strain (Jolivalt et al. 2005 AMB 66:450-6) was inoculated in modified PPB liquid medium with different concentrations of glucose, sucrose and molasses. The laccase activity was determined spectrophotometrically in kinetics mode by measuring enzymatic oxidation of ABTS at 420 nm. Results: The Y. lipolytica YL4 strain was able to produce laccase yields of 0.87, 0.52 and 0.56 U/ml in modified PPB liquid medium with 100 gram per liter of glucose, sucrose and molasses as carbon source, respectively. In this study, laccase production was 2.2 to 3.8 fold higher than in previous studies that used 20 gram per liter of glucose in PPB liquid medium. Conclusions: Laccase production can be increased by changing the substrate type and concentration. Sucrose and molasses can be used as cheap and renewable substrates for laccase production.
