چکیده Ultrasound-assisted extraction (UAE) is a fast and effective method that uses ultrasound waves and solvents to extract favorite compounds from plant materials [1]. In present study, extraction of total phenolic content (TPC) using different solvents (Ethanol 70% and methanol 80% v/v) with ultrasound and conventional method from shoot samples of Stevia rebaudiana medicinal plant was conducted. Stevia rebaudiana Bertoni is a medicinal plant belonging to Asteraceae (Compositae) family that create attention mainly due to the existence of bioactive compounds in their leaves such as phenolics [2]. This research was conducted using a completely randomized design (CRD) with three replications involving two factors namely solvent type (with two levels) and ultrasound-assisted extraction (with two levels). The two-way analysis of variance (ANOVA) calculations indicated that the solvent types and ultrasound using influenced significantly the TPC of samples. The total phenolic content was determined quantitatively using the Folin-Ciocalteu reagent (F-C reagent) with Gallic acid as the standard [3]. Overall, ethanol 70% (v/v) with ultrasonic-assisted extraction showed highest yield (159.288 ± 1.079 mg GAE g-1 DW) while methanol 80% without ultrasound treatment showed the lowest extraction yield (53.979 ± 5.466 mg GAE g-1 DW).
