چکیده In this research, a probiotic strain of Limosilactobacillus fermentum, designated as strain M09 (OQ568196) and isolated from traditional Maragheh yogurt, was encapsulated using alginate (A) and carboxymethyl cellulose (C) biopolymers through extrusion method, resulting two coating types: beads with calcium chloride, and slurry without calcium chloride. The encapsulation efficiency exceeded 85 % in all formulations; the alginate-bacteriacarboxymethyl cellulose (ABC) slurry formulation exhibited the highest encapsulation efficiency (89.48 %). Also, the average beads diameter ranged from 3.51 mm–3.67 mm and mechanical stability after 24 h was 82.5 (AB) and 97.5 %(ABC). The viability of the encapsulated bacteria was evaluated under simulated gastrointestinal (SGI) conditions and storage at 4 ◦C, 25 ◦C and 37 ◦C for 120 days, using colony count and MTT assays. Micrographs confirmed intact cells within slurries and beads. Among temperatures, 4 ◦C yielded significantly the highest viability 5.92 Log CFU/mL and 28.37 % (by colony count and MTT assays, respectively) in ABC slurry after 120 days. Following SGI conditions, ABC slurry maintained 4.47 log CFU/mL and 30 % viability, outperforming free bacteria and other formulations. The MTT results aligned with colony count. Overall, the ABC slurry delivery system effectively enhances bioavailability of strain M09, highlighting its potential as a carbohydrate-based vehicle for applications in food and pharmaceutical industries.
